DNA变性后由于维持DNA二级结构的氢键断裂,由双链变为单链导致:

1、溶液粘度降低:双链结构分子量大且相互缠绕,流体阻力大,断裂成单链后阻力小,粘度变小。

2、溶液旋光性发生改变:变性后整个DNA分子的对称性及分子局部的构性改变,使DNA溶液的旋光性发生变化。

3、紫外吸收作用增强:由于双螺旋结构的破坏导致碱基的充分暴露,表现为OD260光吸收增大。
